I have a dsl40cr. I want to run a 16 ohm 1x12 extension cabnet and use the speaker in the combo as well. What speaker output jack do I plug the extension cabnet into?

Internal speaker in your combo is also 16 ohm, so you have to disconnect it from the socket marked 1 x 16 ohm, and connect both speakers to the sockets marked 2 x 16 ohm.
